When there is complete overgrowth of the agar it is difficult to determine CFU/mL. Is there a way around this issue other than reducing the incubation time? I am currently working with 7 days incubation at 27 deg C. Thanks.
I am unable to change the dilution and incubation times as I need to keep these consistent with previous work.
Phil Geis, your suggestion is helpful, thank you. To clarify, by doing counts each day up until day 7, would I determine the count on day 7 based on the amount of colonies present leading up to this day?
For instance, if on day 3 to 5 there are 5 colonies and on day 6 and 7 there are still the same 5 colonies but much larger in size (complete overgrowth of the agar), I count this as 5 colonies? Whereas if there was an increase in the number of colonies each day up until day 7, I would consider the overgrowth due to a large number of colonies rather than the growth and spreading of a few colonies?
