I'm looking for feedback about saccharide analysis (mono- and di- mainly) from woody plant extracts using ligand exchange HPLC (Ca2+ as counterion: HiPlex Ca column? Others?) and ESLD?
You can also use Aminex columns (in H+ or Ca2+ form) from Biorad. The refractive index detector can also be used instead of ELSD. During sample preparation, the extract needs to be centrifuged to remove solid insoluble material. The extract also needs to be filtered through 0.45 micron filter to clarify the sample. Some times you need to add some stabilisers in the sample before analysing depending on the sample composition.
Thanks for your answer. I'm interested about your experience with that (I understand you use it..). Are you satisfied? I was wondering about H+ or Ca2+ column as we should have organic acids in the sample as well. Any advice? Do you clean your samples on SPE cartridge? Thanks again
H+ would be better as your samples have organic acids in it. You can use sulphuric acid during sample preparation to satbilise the acids. The guard column can be put before the analytical column to clean the samples.
