Hi everyone. I was looking for an expert opinion on exosome analysis. During the NTA, once you run the 100nm standard beads, do you change the camera level and/or focus for analysis of the sample for better clarity? Or the camera setting should not be changed for the sample once it is fixed with the standard? Expert opinion/discussion is appreciated.

The focus should not be changed as focus adjustement is one of the reasons to measure standard beads. Camera settings like sensitivity or shutter can be changed depending on the sample, eg. if the sample contains very small particles it makes sense to increase sensitivity, otherwise some particles might be too dim as the refracted light intensity could be quite low.
