I have made electrospun fibers made by PVA and I want to control over the encasulated drug inside by considering any type of physical or chemical croslinking. Do you have any clue?
You can try to Dissolve PVA in water as required concentration with (5-10) % (w/w) of gluteraldehyde with heating at 60 °C with stirring for an hour then cool the solution and add the drug to be loaded then make electrospun fibers.
Artir, do you think there is any kind of chemical reactions will be occurred between the aldehyde groups and the OHs at 60. If so, it requires adding Aluminum sulfate as catalyst and to raise temp to up to 130 or more to achieve the hemiactal linkages. This is very harmful for drug and of course for encapsulated bio-molecules.
If you can explain your approach little be more that would be helpful.
Ahmed Nada, There are several ways to crosslink electrospun PVA.
For physical cross linking, you can easily attain crosslinked fibers by simply heating the fiber mats at elevated temperature (80C is enough, you can use higher temperature as well). You will find many papers regarding heat treated crosslinking of PVA. You can choose your treatment time and temperature by studying few papers and giving some trails.
You can use gluteraldehyde vapor although I don't think GA should be used for drug release applications. I have seen some studies using epichlorohydrin to crosslink PVA as well.
You can use UV light to crosslinking as well. This is also easy like the heat treatment and you have to find an optimized treatment time, UV light wattage, wave length.
