No "verification" is possible by using only retention time. Three different orthogonal methods are needed for any reliable ID. Adding full UV spectral data for comparison to the pure standard will add another dimension of data and is wise (more scientific). To do so: Buy a true standard for comparison to your sample. The std and sample must be analyzed in the exact same solution for test to be valid. Use the same validated method of analysis. Make sure the std compound is fully retained and resolved on the column (proper K prime, good peak shape etc), then collect a full spectra at multiple points across the peak (20 scans across the peak is good to start with). Turn off any "reference wavelength" software features when doing this. The final result can be saved as an example library spectra for the specific method. Compare the sample spectral data obtained to the data from the injection of the pure std. Note: "UV Libraries" are of little value in HPLC. No two instruments will show the same exact data and any differences in mobile phase, detectors, solutions used and/or concentration may change the collected UV/VIS signal data so overlays are not reliable (only applicable to the specific method/column/instrument/day etc you used, and nothing else). We do not use library spectra obtained by HPLC for comparison (some value, on the same exact instrument, used with the same exact methods they can be better than nothing, as a guide to determine it the two compounds are similar in their UV/VIS spectra).

Terminology: A 'Calibration' and 'Spectra' have nothing to do with one another. Calibration for quantitation should utilize at least 5 different levels (conc) per order of magnitude. "Reference Spectra" have different definitions for different software packages so please be careful as the incorrect meaning and use can fully invalidate the analysis (e.g. Reference Spectra could be defined as "Spectral plot of a pure compound" or some define it as baseline spectra used as the reference to compare the spectra to).

The apex spectrum of "ANY" Peak ? That makes no sense at all. A reference can only be one thing, not many. Sounds like something is taken out-of-context here or the manual is rubbish.

I am no native speaker (sorry for that, indeed). Maybe the author of the manual is also a German and yes, there are typical errors when Germans use the English language. What is meant is obvious for a non-native speaker like me: you can choose every single peak you want as source for the reference spectra ...
