i'm trying to detect the natural luminescence of some bacteria. I'm using white frame and clear bottom plate (perkinelmer Isoplate-96 TC, Tissue Culture Treated, White, clear bottom, tissue culture treated, sterile, with lid, 96-well) to detect ABS first and then Luminescence sealing the bottom of the plate with the white backseal.
when i read the luminescence in a time course experiment, it achied a maximum of ~7log CPS and i'm quite happy about it. However, the signal blank seems to be quite high variing from 500 to 100. Few month ago i recorded it at abou 50 CPS.
The protocol ued is: 10 sec orbital shake at 100 rpm, detector 0.1 mm, time reading per well 2 sec,
I optimized the plate but i do not find very clear the instructions.
Anyone use this model and have experience with luminesent bacteria?
