The quedtion aims to seek help on information or literature on how to quantify the amount of metabolite(s) in a crude plant extract of dried Calotropis procera leaves.
I wish to determine by volume each of saponnins, tannins etc. in the plant extract.

However, the plant chemical composition is highly variable due to several factors (e.g. soil, climate, etc) and on the type of extraction.

By doing GC-MS analysis You have to quantify the quantity of saponins and tannins compounss
By precipitating the particular metabolite with the appropriate precipitating agent, for example saponins may be obtained by extracting the plant sample with n-butanol after removing the fatty matter with n-hexane and then precipitating the saponins in the extract with diethyl ether. The dried precipitate can then be weighed and taken as amount of crude saponin in the sample. alkaloids, flavonoids, tannins etc can be obtained using the appropriate ppt agents.
You can quantify secondary metabolites by HPTLC, HPLC, GCMS and LCMS etc. modern techniques and you can by precipitate by appropriate precipitating agent also.
The method depends of the metabolites you want to quantify.
The bests are GC-MS or HPLC. However many others exist
It is very difficult to qualify plant extract. In my opinion, select the plant extract responsible for desired activity and quantify that in extract by appropriate method
