I need a calibration curve of curcumin on UV- spectroscopy, but in several articles have been reported that we should be used PBS - PH=7/4 to calculate releasing of curcumin from the scaffold. This is while curcumin is not soluble in PBS (or is very low solubility ). How do I provide a calibration curve of curcumin for UV- spectroscopy?

You can go to lower concentration (10 (in an apropriate wbuffer) hen the solubility is higher.
You can dissolve curcumin in the lowest amount of organic solvent (eg. DMSO, carboxymethyl cellulose etc.) and make up the volume using PBS. (you can use the same dilution mixture as blank for spectroscopy)
I totally agree with Nigam. I would like to add the following, normally we use concentrations of 1-25 micro gram to obtain a calibration curve absorbance
The problem of curcumin is the stability in methanol or water as solution.
After 30 min of solution preparation it loss about 90%.
Use MeOH containing 0.1% ascorbic acid.
But this solution could be analyzed by HPLC.
For UV measurement, you should make sure that the measuring wavelength selected for curcumin (390 nm) is not interfere with ascorbic acid, in case of UV measurement.
