Can anyone please provide me CLSI document M27-A3 ? I want to perform broth microdilution method for antifungal susceptibilty testing but do not know how to do it..Can anyone please explain it from the beginning in a simple language?
PROTOCOL:
· Label the test tubes from 0 ,1st ,2nd …….8th
· Add 2ml media to each tube, 4ml to 1s t tube.
· Add desired concentration of test compound to tube 1st only. Mix solution for 2 minutes and transfer 2 ml to tube 2nd.
· Transfer 2ml again from tube 2nd after vigorous mixing to the tube 3rd. carry out serial dilution (by transferring 2ml of mixed) till 8th test tube.
· Discard 2ml from the 8th tube.
· Add culture according to McFarland Standard at O.D 0.1 at 625 nm
· Incubate tubes in shaking shaker at 37ºC 150 rpm for 24 hours.
· The resulting visually clear tube would be MIC of the test compound.
