Dear friends,
I am working on combination of two modalities which I believe to promote synergistic effect. I only found one paper that mentioned the equation to measure the interaction of combined therapy (which is secondary source). I attached the source for discussion. What make me unsure is that they seemed likely to multiple of each individual modality rather than the sum value among of them. Could anyone help me for more detail? I hope you provide the sources too if it is possible, thank you.

Isoblographic analysis is what I recommend for your study. It is robust and allows you measure antagonistic, additive or synergistic effect of combinations. The isoboles will give you a pictorial look at the interaction while the interaction index will estimate the degree of the observed interaction. Note that statistical comparison of the theoretical and experimental potency is important for conclusive deductions.

Synergism and its effect as a basic rule of thumb is when they amplified in their effect, as they belong to the same group of drugs!
In my clinical experience every synergist when administered in combination may be reduced to a dose of from 1/3 to 1/2 less than its Referential doses, and the obtained cumulative better therapeutic effect, and reduces the side effects of the drug when administered in monotherapy!
I forgot one important thing to note on that particular attention should be paid: that there are cases in clinical pharmacology fact rarely or there that the joint effect of the synergist is greater than the sum of their individual effects of this type supraditivnog synergism is very difficult to reliably in two synergistic drug .
More commonly, the drug, which is in itself inactive to stimulate • increases the effect of the active drug!
