What do you mean by "antioxidant activity of blood"? Do you mean total antioxidant status? If yes, it is normally assayed in serum or plasma not whole blood. However, you can assay SOD, CAT, GPx and GR in whole blood. They are many available commercial kits for them.
You can measure Total Antioxidant Status (TAS) using commercial kits (i used from Randox), or you can carry out Ferrous Reducing Antioxidant Potential (FRAP) assay.
An interesting method of analysing AO activity of your drugs/extracts could be by investigating its ability to inhibit APPH-induced hemolysis. This is something i have done and published. You can use whole blood for this or serum.
thanks to all for for ur precious suggestion, i have idea about using kits for AO assays but due to financial constrains i want to use spectrophotometer for AO assay instead of commercially available kits.
My experience says that there is no significant difference in the cost of kit and manual method, otherwise, you have the materials needed for assay (you don't need to by the materials). Also, kits are ready to use and optimized.
