I would like to know if my lncRNA is polyadenylated. To investigate this, I conducted an experiment using the same samples for generating cDNA using random primers in one set and oligo(dT) primers in another. Subsequently, I performed qPCR with a housekeeping gene, GAPDH, and my lncRNA. The results showed Ct values of 18 for GAPDH with random primers and 23 with oligo(dT) primers. However, for my lncRNA, the Ct values were 27 with random primers and 25 with oligo(dT) primers. I'm unsure how to interpret these findings. Any insights or suggestions would be greatly appreciated.
