My crayfish antennule cryosections keep falling off my Fisherbrand Superfrost Plus Microscope Slides slides when I wash them (when I lay them flat or in coplan jars). I can't use my IHC protocol because the sections will not stay on the slides at all. I have ordered new slides to see if that is the problem. I have never sectioned antennules before. Is there a problem with the chitin sticking to the slides? Has anyone tried an adhesive that worked?
Suggestions are welcome!
U can try Poly-L-Lysine or gelatin coated slides. As I hv been using during my doctorates, worked well with fish brain sections. As per my experience Poly-L-Lysine works well than gelatin. And as u texted presence of chitin could be one of the problems as it is much stiff in nature and can interfere with the bondings.
Buen día Rachelle: nosotros solucionamos el problema poniendo una delgada capa de Albumina de Mayer en el porta limpio antes de recoger el corte. La formula para prepararla es:
- Disolver 1gr de ovoalbúmina en 100 ml de agua destilada, añadir 100 ml de glicerina. Filtrar y añadir disuelto en algunas gotas de agua 1gr de Salicilato Sódico ó Timol.
Con un pincel poner una pequeña cantidad en el extremo del porta y extender con el dedo. Una vez colocado el corte pasar el porta unos segundos por una plancha térmica para que el corte se estire y coagule el albumina, esto hace que el corte se pegue al porta y no se desprenda durante la coloración.
