My kit recommended generating a four parameter logistic (4-PL) curve-fit for my standards; however, I can't find any tutorials for how to do that on GraphPad. I did find a tutorial for generating a second order polynomial (quadratic) fit and I was able to analyze my data and determine concentrations of my samples with it. I'm a total novice at ELISA so I'm unsure why one would be preferred over the other! I am interested in learning how to do the 4PL fit so that I can compare which standard curve is a better fit for my data and how they affect my concentrations.

Any help or direction would be much appreciated!

The kit I am using is a Quantikine ELISA Human GDF-15 Immunoassay
