Does someone have a detailed protocol for Proteoglycan extraction from ocular tissue such as cornea or sclera? or have any experience with PG extraction? List of supplies and chemicals are also needed. thank you
Hot alcoholic extraction can extract glycans, and cold acetone and alkali can extract proteins. Actually both are intermilgled very tightly hence, both can not be extracted in one. Similarly, separate extraction should be done for carbohydrate as well as proteins
I read some papers in which the researchers used Guanidine HCL to resolve the proteins and then dialyzed the extraction into UREA, following that DEAE columan was used to elute the PGs. But I need a protocol with detailed procedures and comments.
