I would like to know if I it is possible detect H2O2 in Transmission electron microscopy through DAB and osmium tetroxide reaction. Doe anyone know if it is a possible to detect hydrogen peroxide by stain it in bloco and then observe on TEM?
"using the cerium chloride technique which leads to the formation of per- hydroxide precipitates in presence of H2O2, was performed to localize H2O2"
"Leaf pieces were immersed in this incu- bating solution under vacuum to allow a rapid penetration of CeCl3 into plant cells. After four washes in 50 mM MOPS buffer, cells or plant tissues were fixed in a mixture of 100 mM sodium cacodylate buffer (pH 7.2) containing 1.25% (vol/vol) glutaraldehyde and 1.25% (wt/vol) paraformaldehyde for 1 h at room temperature. After fixation, samples were washed sev-eral times in the same buffer over a period of 30 min"
It was a great! Thank you for shed light on this topic.
