I am trying to set up my next experiment, but I am not confident I correctly understand the basic principle behind spontaneous immunoglobulin production.

How can PBMCs from healthy humans, when cultured in vitro (for example in 96-well plates) and without any stimulants, produce antibodies? Is it because of a cell subpopulation that is pre-activated? And according to the protocols, why does it take 7 days for cells to produce antibodies? Is it for B-cell differentiation to plasma cells? But once again, what is the differentiation trigger?

Could this be also the reason for the elevated basal expression of AICDA gene (5 fold via qPCR), that I found in my PBMCs, 72 h after the start of incubation? Are the B cells turning into plasma cells?

PBMCs were cultured in RPMI-1640, with only 5% charcoal-stripped FBS.
