You can use optical microscope (reflection mode) to quantify the layers of graphene by simple method.

Simply coat it on the substrate and observe it through OM.

Its shows different colors with increase in layers.

Rhys, can you please detail your answer? Infact Raman is not differentiating graphite-graphene mixture, especially when graphene is a nanoplatelet (>15layers).
