For my project, I need to cap synthetically prepared RNA oligos and most of the capping kits available require a triphosphate group at the 5' end as a substrate. I have looked all over for kits and enzyme protocols to do this. I have so far found the 'Ice and Fire' phosphorylation kit (http://www.prokazyme.com/index.php/products/polynucleotide-kinases/50-ice-and-fire-phosphorylation-kit.html) but it is rather expensive. I have found T4 Polynucleotide Kinase suppliers. Will that enzyme or any other be able to produce a triphosphate on the 5' ends of RNA oligos (and not just monophosphate)?
