In my experiment, I want to know the percentage of naive and effector/memory Cd8-t cell in mouse spleen by FACS. As I understand it, naive Cd8 T cells have high expression of CD62, effector/memory Cd8 T cells express CD44 highly. Can I stain spleenocytes with CD44/CD8 or Cd62/Cd8 to distinguish these cell populations?
And how can I identify CD44 high/ Cd62 high by FACS?

Depends how many colours you have available. I found using CD8 instead of CD62L worked for me according to Lanzavecchia's strategy. It's published in T cells reactive with HLA-A*0201 peptides from the histone demethylase JARID1B are found in the circulation of breast cancer patients on my profile if you want full details.

I wonder which FACS machine you are using. If you use BD Calibur, it's really limited because of 1 blue laser. FACS Aria is the better choice with red and even violet laser.
I usually stain CD44 - FITC, CD62L PE, CD8 - PerCPCy5.5, and CD45RA -APC
If you just need to distinguish naive and memory/effector, CD8 and CD44 is enough. CD8+ shows very clear 2 populations: CD44high is memory/eff and CD44low is naive.

I am analyzing blood from mice and I found a population CD62L-CD44- gating both CD8 and CD4 T cells. Do you find them as well? If they are not naive, nor effector or memory, what are they? I would really appreciate your thoughts!
