I am currently working on testing the fitness of E. necator, an obligate biotrophic fungi that attacks grape vines. My lab believes a fungicide resistance mutation may have an energy cost and that this may effect the reproductive fitness of the pathogen. I have a few ideas from the literature but I would love some community experience/ideas on how to test obligate biotrophs for fitness.

If your mutation for fungicide resistance is affecting the fitness of the pathogen this should be measurable in assay of sporulation capacity of the wild type compared to mutated type when both have no fungicide applied. Wild type sporulation minus the mutated type would be fitness delta value. If this difference delta value was divided by the original wild type sporulation and multiplied by 100 the value would represent the loss of fitness from the mutation. The replicated values of the mutated and wild types can be used for testing the hypothesis that the values are different and the probability that they are different. The testing of the wild types and mutated at different fungicide rates would also be insightful. Hope you find these thought helpful.
