I have good staining of anti CD3 and CD8 antibodies but they do not colocalize in mouse intestine cryosections fixated with paraformaldehyde. Do you know how to solve this?
It would be a good idea to look at the pictures since CD3 is a general T-cell marker so certainly many more cells will be positive with this marker than with CD8+ which will stain is a smaller subset. Are you doing immunofluorescence or peroxidase-amplification with stains visible in brightfield? -
Only a fraction of CD3+ cells will be colocalize with CD8+ but not all. Try confocal with maximum projection in case you care unable to find that with wide field.
