Do you want to separate the polar compound for analytical purpose or synthetic / preparative purpose?

What is the matrix, quantity of sample/ bulk?

If you want to extract polar compounds then you can try mixtures of ethyl acetate and DCM. If this combination doesnt work then choose solvents with low polarity index than ethyl acetate.And chromatographic sepration of polar compound can be achived by using reverse phased chromatography,use C18 columns or you can use HILIIC columns also.

Its dosnt matter. we know the basic principle about polarity of solute depands dielectric constant with specific temperatures. Once we know the dielectric contant of known solute then we could choice as you priscribed of anyone method. example water dielectric constant (DEC) 80, but it is protic solvent. Hexane is non polar solvent, Ethyl acetate and DCM are aprotic solvents and them DEC around 6 and 9 respectively. by choicing of these solevents should be equal to solute polarity, that help us to better yield of product selectivity. Sometimes the polarity lies between what we mentioned early, that we can adjusting by mixing of two differentsolvents for required one.

i understand, you are going to use preparative HPLC. use any polar column (RP amide....), methanol can be used as solvent.

HILIC could work, if not then use ANP columns. Such as diamond hydride columns:

These columns are really good. Other options include normal phase chromatography or SFC.
