I have two questions to ask
1)-I would like to change some amino acids from my enzyme(wild) to achieve particular function, but I do not know how to select and substitute my enzyme’s amino acids, is there any software to use ?
2)- How can I compare structure of my wild enzyme and engineered enzyme ?Thanks
Hi Ali, if your enzyme is not to big, then you could syntheses it with the amiaocids of your choice.
You mentioned the word "software", so does this mean you are attempting theoretical calculations only or do you want to synthesize a modified protein?
I agree with Khaleel. Software can only give you a virtual reality of your synthetic protein. Making the protein will give you actual data of its properties.

At first I want to find the candidate amino acids to change (by appropriate software) and then I will experiment my substitutions on bench , but I don't know which software is suitable to use
You need to decide which amino acids you want to introduce. For example, if you want to replace a metal-binding amino acid with one that does not or replace a hydrophilic residue with a hydrophobic one to see the effect on the tertiary structure or something else.
There are so many possibilities out there, but first you should decide the goal of the change you want to make.
