I want to demonstrate the result of segmentation by reveal gray scale image and draw two color lines one denotes the result of my algorithm and the second denotes the ground truth
what you are using ? i would copy the grayscale into an RGB image (R=G=B = G for 8bit do the scaling for other ... some libraries do so) and then i may add what ever i want in any color. (ImageJ is nice)

i use rarely Matlab, however i believe what you want is saving the overlay mask (probably Alphamask). google led me there which seams interesting:

The resulting image is not more gray scale image.
Let suppose that you have the grayscale image Ig and the contour Ic of the same size.
- Convert Ig to RGB (replace R band, G band and B band by Ig); let call this converted RGB image Img.
- In each RGB band of Img, replace all pixels of contour by its color number (for example (225,35,10) for (R,G,B) band) before display Img image.
