I am working on mouse liver lymphocytes to study the immune response against schistosmiasis but I am having difficulty getting pure single cell suspension because the mice that I am now working on are schistosomiasis infected and the parasite eggs are normally accumulated in the liver of the mice. And it was difficult to analyse the flow cytometry data because of the eggs. Is there anyone who can advice me what I should use to get rid of the eggs from the single cell suspension of the liver lymphocyte ?

Suspend liver homogenates in 5 mL of 1xPBS and load it in to percoll-sucrose gradient consisting of 8 mL of percoll and 32 mL of 250mM sucrose solution. Then Eggs will be separated from liver cells by centrifuging at 800xg for 12 minutes. Take the supernatants which is your cells and discard the egg pellets.
