I want to making Andrographolide nanoparticles (Andrographolide (AG) is a herbal lipid) In order to encapsulation of AG in PLGA and make nanocapsules I have done the following steps: 1) 50 mg of PLGA and 2.5 mg of AG were dissolved in 3 ml of chloroform. 2) 4% aqueous PVA (98-99% hydrolyzed) was added as an emulsifier to provided solution in step one. 3) The solution was magnetic stirrer for 12 h, sonication for 3 min , homogenization for 20 min and centrifugation .The problem is that although the 4% PVA exist, but the AG-PLGA precipitates out or attaches to the wall of the beaker. Can anyone please let me know, what could be the problem? And what measures should I be taking? Is there any other proper emulsifier or stabilizer for my experiment?
