Yes, I have observed the same thing. When there is enough bio-reducer there is no formation of nanoparticles, and if there is excess bio-reducer increases the particle size. I tried the synthesis of silver nanoparticles with Hetheroteca inuloides, with green tea and clove, it appears that the optimum concentration is 1 g of extract per 100 ml of distilled water.
A convenient way to monitoring the concentrations to be tested is using uv-vis analysis each hour.
You may need to run many trials with different concentrations and monitor formation of nanoparticles by any suitable mean i.e. size (zetasizer), shape (SEM) or absorbance shift (spectrophotometer). Zetasizer and some spectrophotometer have kinetic mode through which you can measure the formation of nanoparticles over time.
You can also use a statistical design (such as Box-Behnken design) which will show effect and optimum concentration of each component for desired nanoparticles.
