Open your 3 images for the 3 channels in Photoshop. Copy two of them into the third - select all (ctrl+A in windows) and copy (ctrl+C) and paste it (ctrl+V) into the other file.

Now you will have your 3 channels as layers. Go to the "Layers" pane and double click the the top layer (you can rename it if you want by dbl clicking the name of the layer). Now you get to the "Layer Style" dialogue. Under "Advanced Blending", uncheck all boxes except the color of the channel.

Do the same for the middle layer. Now you should see your channels merged.

just use ImageJ. it's a lot easier.

You should appreciate the fact both approach might have similar applications but mechanism are different. Take a picture wash with fluorescence dye and develop using immunofluoresence then you can upload on photoshop. Image appears sharper and alive..

I found Marina C M Franck's explanation to work perfectly for merging my coloured channel images (red, green, blue), however, I cannot achieve the same for a channel that is "white", which in my case is stained with fluorescent secondary 647 a "far red".

Now when I try and perform these commands, any part of the image that is overplayed with colour becomes yellow.

I can't find an answer anywhere on the internet and I've tried many different blending options. Does anyone know how to merge a multichannel image containing a "white" channel?
