I don't quite follow what you've written. As you mention, Indo-1 is used as a dual emission indicator. You need a single excitation source set to ~340 nm. Your PMTs will be measuring the output from Indo-1. One should be set to monitor light emitted at ~405 nm, and the other ~490 nm. The ratio of light recorded by the PMTs (i.e. the ratio of the 405/490 signals) is proportional to calcium.

So, you shouldn't have both PMTs on 340 nm: that's the excitation wavelength you need. The PMTs need to be set to record light of different wavelengths as above.

Sorry for the confusion, but I believe I have set up my PMTs just as you have mentioned. I just want to know how to see both emissions on clampex given that I only excite with a single wavelength. I have no experience with clampex so forgive me for asking such a basic question. Thank you.

You may simply use AxoGraph and MiniDigi-1A to acquire digitized data from PMTs.

I used liquid crystalline to measure of DSC and POM. Sorry
