I am tirying to feed the rat with rat chow supplemented with 2% cholesterol, 1% cholic acid for making hypercholesterolemic rat model. However, it is difficult to dissolve cholic acid. As the last resort, I sprinkle cholesterol and cholic acid in the ethyl ether on the chow. Thus, I would like know how I make rat chow with cholesterol and cholic acid.
You may purchase commercially prepared atherogenic diets, or maybe you can prepare the diet yourself? If you find your desired recipe you must blend the ingredients and make pellets or flakes (dry by moderate heating), the more calories (fat and sugar) and cholesterol in the feed (ie. resembling a Western-type diet), the more cholesterol will accumulate in the rat serum.
try a high fat feed containing 4 kcal% fat (lard), 35% carbohydrate (corn starch, sucrose, and maltodextrin) and 20% protein (casein). feed the animal for at least 5 weeks and check out the results.

I agree with Dr. Eilertsen and Dr. Balogun. Or you can add Ricotta Cheese or better yet
salty Feta Cheese in the diet you already feeding the animals. In 2 weeks the resutls will be pretty fatty.

If your budget and importance of your research allows it you can also consider transgenic or KO rats. Several companies offer the service these days. They use the CRISPR/Cas9 technology to obtain animals in a few month. I have not used the service but I am considering it for mice. As in mice you could try the transgenic plus the high fat diet .
