I tried to make diabetic rat using streptozotocin; After fasting for 24 h, streptozotocin (STZ; 65 mg kg−1) in sterile saline was injected to Sprague-Dawley rats (300–350 g) intraperitoneally. However, rats died with convulsion overnight. I wonder why did the rat samples die and animals should be fasted for a further 24 h after injection? I should be very grateful to you if you might help me
Streptozotocin induced diabetes provides a relevant example of endogenous chronic oxidative stress due to the resulting hyperglycemia. STZ is a pancreatic cell toxin that
induces rapid and irreversible necrosis of cells.Whereas a single diabetogenic dose of STZ (70-250 mg/kg, body weight) has been demonstrated to induce complete destruction of cells in most species within 24 hour, multiple sub-diabetogenic doses of STZ partially damage islets, thereby triggering an inflammatory process leading to macrophage and subsequent lymphocyte infiltration, which is followed by the onset of insulin deficiency.
Streptozotocin causes sudden, massive and irreversible destruction of pancreatic beta cells and causes diabetic coma followed by death. Dosage standardization is very important with STZ induction. It has to be done at one to two animals at the beginning instead administering for a group of animals.

Hi, we have used very successfully , a single dose of 65 mg/kg STZ i.p. in ph 5.5 citric acid buffer with no fasting, in 300-400g male rats. We allow them access to a 2% sucrose solution for 48 hrs following STZ, this is in addition to their normal drinking water. We also allow an equilibration of the STZ solution of ~ 30 mins prior to dosing. Hope this helps....
I advise to inject a lower dose of STZ (6-10 mg kg-1) for several days, 7-10 days, controlling regularly glycemia. The Accuckeck strips are enough. You will see that after two weeks starting treatment, animals will have blood glucose levels over 300-400 mg / dL at baseline associated with significant weight loss that will be very important after 3-4 weeks. To confirm pancreatic destruction will require a quantification of basal and postmeal insulin. Ideally, if you may, quantify C Peptide . It is very important the water supply because the diuresis of animals will be much higher. Similarly take care to clean boxes regularly.
Once established diabetes, have to start daily insulin administration protocol which should suit your needs. Best.
