There are many researchers said the size of reactive astrocyte is larger than normal astrocyte. But how can I judge the reactive astrocyte by GFAP or S100beta staining? For example, what size of astrocyte (> ? μm) can be regarded as activated astrocytes, or some other index can be considered.

Redefining the concept of reactive astrocytes as cells that remain within their unique domains upon reaction to injury by Wilhelmsson et al.
In it they reveal a slight increase in the overall astrocyte size, however the main indicators of reactive vs unreactive astrocytes are an increase in GFAP immunoreactivity that can be observed as an increase in apparent soma size and thickening of processes especially proximal to the soma. Relative GFAP expression can be observed/measured using non-amplifying/direct immunofluorescent detection during immunohistochemistry.
