I want to isolate murine macrophages from the tissue surrounding ligated nerves using MACS, and was told by the MACS rep that since there are no microbeads for F4/80, I would have to work indirectly using anti F4/80-PE (or FITC, APC, PE-Vio770, PerCP-Vio700, Biotin) and then using anti-PE (or FITC.. etc) microbeads. The problem is that after I have isolated these cells, I want to carry out FACS where I already have antibodies conjugated with APC, PE and efluor 450. Which would be the best MACS combination to use? Will it interefer with the FACS experiments?
