I have to isolate some microbes having role in hemicellulose and lignin degradation. How can i isolates that specific microbes for hemicellulose and lignin degradation on specific media. Which types of media will be best for isolation and screening of targeted microbes as hemicellulose and lignin degrader.

I would use media with complex carbon sources such as xylan or soluble cellulose, since simple sugars tend to repress ligninolysis. You can also add enzyme substrates that reveal ligninase activity, such as guaiacol or azure B so you can visualize and select colonies. Congo red is helpful to see hydrolase activities such as cellulase, but since you add it after the organisms grow and it is a destructive method, you need to transfer the colonies before performing it, so I would leave it for a second step.
