Collect any soil samples from a core below a depth of 15 cm, and place sub-sample into a phosphate buffer or saline phosphate buffer or even filtered ambient water. Homogenize the material followed by serial dilution. Then inoculate from the most diluted batches. At this stage use a media used for fecal coliforms such as Tryptose lauryl sulfate broth. Following incubation at 37-42oC in a sealed vial you'll notice black precipitates at the bottom of the container, these are nothing but FeS produced by the reduced S with the Fe in the media or in the environment. Then you'll be sure of the presence of SRBs.

Following this all you need is to get you hands on a media like the Postgate's modified B and you can inoculate the bacteria from the vial to isolate the non-fastidious an cultivable SRB strains easily, in my humble opinion.
