I have recently started working with SF9 cells and they have problems attaching to the TC flasks with less than 40% attached. I have also tried using poly-D-Lysine with little effect. In my mamalian cell culture work I use rat tail collagen coated flasks for cells that struggle to adhere, can I also use this for my insect cells and what are some of the advantages and disadvantages of using this over poly-D-Lysine?
My experience is that insect cells attach well when they are healthy and happy. How are you growing them in general? You should grow them in a suspension culture and only seed them for transfection.
They are recent startups, and I am growing them as monolayers in TC25 flasks with TC100 media.
I'll keep your suggestion of growing them in suspension in mind and discus it with my supervisors.
