I am trying to extract the lipids from microalgae by Bligh and dyer method. After performing chloroform, methanol and water bilayer has formed but chloroform containing lipid layer has green color due to presence of chlorophyll. So, how can I get over it. I need your valuable suggestions for this problem
Blight and Dayer method as far as I know, for determining the crude lipid content including ( Triglycerides(TG), free fatty acids, phospholipids, lipoproteins ...etc). due to high solvent polarlity used in this method( methanol and Chlorophorm) you will get traces of pigments content with your samples ! any way this article has a crucial answer for your quest

Hi Ashutosh. Have you tried washing your isolated chloroform layer with chlorophyll pigments with saturated NaCl in water? Perhaps give it a try on a test sample if none of the other methods work for you.
