Are you using polar birefrigence? if so, you need to adjust the microscope only. If you check only with light microscopy, you will not get this yellow colour out.

I'm using light microscope but in the same field l have yellow area and grayish areas so when I use image j to calculate the fibrosis percent I have wrong results

To calculate fibrosis percent, you better use a filter of birefrigence. With this you can even calculate the area from different collagen types and thickness.
