I am in the process of formulating an oral dosage regimen for olorofim (HY-104029) that ensures safety, non-toxicity, and improved solubility. The intended dose is 16 mg/kg for an average animal weight of 10 kg, with a gavage volume of 20 mL, resulting in a calculated working concentration of 8 mg/mL.

I am considering several dissolution regimens and would like to know if they are feasible:

  • 0.5% CMC-Na
  • 50% PEG 300 + 50% Saline
  • Hydroxypropyl methylcellulose acetate succinate formulation (HPMCAS)
  • Hypromellose acetate succinate-based formulation (HPMC)
  • If anyone has experience with these formulations or can provide insights into their viability for this application, I would greatly appreciate your input!
