For Nematodes, molecular identification is easy and you can perform at your Lab. You can use regions of Taxonomic importance like ITS rDNA, 28S gene or 12S gene for their valid identification up to species level. But you need to have PCR and the material required for doing all this.

Aashaq Hussain Bhat, but that does only apply to single nematodes does it? Or can I extract DNA from all nematodes mixed, perform a PCR with Primers for the gene regions you mentioned and then... then I need NGS, don't I? Because sequencing and barcoding would be futile with so many different species in one sample with conventional sequencing?

I was just wondering if you could identify the nemas to species level correctly with only molecular methods. I am sorry but many species with false identity have been found deposited by some of the molecular taxonomists as they had no idea of the morphology of the species they were dealing with.

Mohammad Mahamood , as far as I've come with this so far I totally agree. The only results in NCBI you can trust are nematodes with quarantine status. Clear Detections sells a hand full of species specific kits, but financially it's impossible to do this for every species you think possible... So far we tried single nematode assays and if already this sends you to relatively unspecific NCBI and BOLD results you can imagine how the results for 100 specimens at a time will be evaluable.
