I am using Tris phenol for extraction of protein from a plant sample...at the end of the extraction process I am unable to dissolve the obtained pellet in buffer(tris hcl) by using vortex. But I need to dissolve the pellet for gel loading ... how can I dissolve the pellet, please suggest.
The protein is completely denatured by phenol. You will have to use a solubilizing agent such as an ionic detergent, or urea or guanidine HCl to dissolve it. If you are going to perform SDS-PAGE on the dissolved protein, you might first try to dissolve it in SDS-PAGE sample buffer.
