I agree with Claretta, In cryostat section you can detect various lipids from unsaturated ones to  phospholipids.

Thanks Claretta and Angella for your answers, but actually I want to destroy the total tissue lipid content in a FFPE section and want to check the treatment effects further. So any suggestions for lipid destruction and quantification techniques keeping other nucleic acid or protein content as it  is?

Dear Saratupa! You must remember that histological processing of biological specimens to obtain paraffin-tissue sections include the use of solvents (ethanol, xylene) and temperature near to 56 oC to paraffin infiltration. These procedures eliminate many lipids, so will be difficult to stain it. Once you obtain tissue sections on a tissue slide try a acetone treatment or polyethylene glycol in several concentration and several times and probe Oil Red-O, SUdan stain and Osmium tetroxide. Treatment of sections with a mixture of chloroform+ethanol (3:1) for about 5 min may rid your sample of the lipids. When lipid is extracted, we expect all membranes are destroyed which compartmentalize cellular organelles and cells. It is not clear, what is the purpose of doing this?
