I performed ethanol extraction using 50 g of Nigella sativa with 250 ml ethanol and 100 g of turmeric with 500 ml ethanol. After filtration, we proceeded directly to nanoformulation without evaporating the ethanol solvent. The nanoformulation was prepared as follows:

  • Stock solution: 3 ml acetic acid + 300 ml distilled water
  • TPP solution: 0.05 g TPP + 50 ml distilled water (for each formulation)
  • Chitosan + Turmeric Nanoparticles:0.5 g chitosan + 100 ml acetic acid stock solution (stirred for 20 min) 20 ml turmeric extract added dropwise while stirring (no heat) 50 ml TPP solution added dropwise
  • Chitosan + Nigella Nanoparticles:0.5 g chitosan + 100 ml acetic acid stock solution (stirred for 20 min) 20 ml Nigella extract added dropwise while stirring (no heat) 50 ml TPP solution added dropwise
  • Chitosan + Nigella + Turmeric Nanoparticles:0.5 g chitosan + 100 ml acetic acid stock solution (stirred for 20 min) 10 ml turmeric extract + 10 ml Nigella extract added dropwise while stirring (no heat) 50 ml TPP solution added dropwise
  • Since we did not evaporate the ethanol solvent before nanoformulation, how can I determine the concentration of crude extract in each nanoformulation to proceed with the MTT assay?"
