I want to do a sample recirculation with ÄKTA system and sample loading pump---50 ml sample goes through column installed in the ÄKTA system and then goes back to the sample beaker. During the sample recirculation, I hope there is no buffer through the column, only the sample.
I create a automatic method as followed,

0.00 SampleFlow_960 2.0 {ml/min}

0.00 Wavelength 280 {nm} 254 {nm} OFF {nm}
0.00 DirectLoad_960 2.0 {ml/min} 100.0 {ml}
4.00 End_Block
However, there is some problems,
1. “Directload” will sample go through the whole system and column? I found it was staying “hold” mode.
2.With this method, I can not read the pressure of system.
3.Does the “Flow” only mean the flow of buffer, not the sample? Because, if I did not set the “Flow” as 0.00, the buffer would also go through at the same time.
If anyone had related experience or any suggestion, pls let me know.
