You'll have to test also ascorbic acid, make a standard line of ascrobic acid in the DPPH assay and from that calculate the concentration of ascorbic acid that gives IC50; As DPPH is not very well standardized, you can not just find it in the litt.
You cannot, but to calculate the results as mg VCE/ g of sample you must use different concentrations of vitamin C with DPPH solution, and use the relationship between the concentration of vitamin C and absorbance to fined the equation that use to calculate the antioxidant activity of extract (equation + the absorbance of extract) as equivalent milligrams of vitamin C . That means Equivalent milligrams of the vitamin, which gives the same activity
Good day all. I am also having issues with expression of DPPH in mg ascorbic acid/g and the answers provided have not really helped me. I have carried out the necessary extractions and absorbance readings to obtain % inhibition of free radical scavenging activity using [1-{absorbance of sample/absorbance of control}] * 100. I have also plotted the absorbance of ascorbic acid vs concentration of ascorbic acid to obatin IC50. Please what formula, if there exist any can I use to express my answer in mgAA/g. If non exists, what is the best form to present my results?
