Im working on bacteria exposed to salt stress and I don't know how to calculate the osmotic potential of bacteria by osmometry. Does anyone knows the extraction and calculation method?
Solute potential can be calculated in the intact cells, without extraction, as described in the following two publications: 1- Mansour et al (1993) J. Plant Physiol. 142: 623-628.
2- Lee-Stadelmann and Stadelmann (1989) Methods Enzymol. 174: 225-246.
If you want to have an extraction from bacterial cell, you should have sap press and make an extraction, then centrifuge to remove cell debris. determine the osmotic potential of the purified extract using an osmometer. This is briefly for extraction, but I do not remember a publication.
