Compusyn allows us to use a non-constant ratio for combination experiments, but it does not calculate the IC50 for the compounds in combination and only gives the normalized isobologram.

In order to calculate the Combination Index (CI) and plot the Isobologram, we need the IC50 of each individual compound (which I can calculate using Graphpad) and the IC50 of compounds in combination

Is there a way to calculate the IC50 for each compound when in combination when using a non-fixed ratio experimental design?

Example:

Compound A fixed at 1 uM + Compound B 0.5 uM

Compound A fixed at 1 uM + Compound B 0.75 uM

Compound A fixed at 1 uM + Compound B 1.0 uM

Compound A fixed at 2 uM + Compound B 0.5 uM

Compound A fixed at 2 uM + Compound B 0.75 uM

Compound A fixed at 2 uM + Compound B 1.0 uM

I would really appreciate your help! Thanks!!
