We want to isolate urine derived stem cells from voided urine as it has been described by many researchers. Unfortunately, all our cultures are contaminated. We use the standard 1% Pen Strep, but that doesn´t help. Does anyone have a suggestion how to avoid these contaminations or uses other antibiotics? I am thankful for any ideas.
It's not easy the full separation between eukaritic and microbial cells. You can remove a lot of bacteria and viruses using filtration or slow centrifugation and some washing steps. However the sample won't be clean enougjh f microorganisms. You can try with a combination of gentamicin: cloramphenicol , probably it will kill all bacteria however with yeast and fungi also will be needed compounds than can be toxic for your steem cells, you can try with nystatin.

make sure the urine is not contaminated from the beginning. in other words, that patients do not have urinary tract infections. however, the addition of 50 mg / L of chloramphenicol to the culture medium may serve (provided that patients are not treated with).
