How can differentiate plasmacytoid Dendritic cells from bone marrow cells with grow factor with good yield?

I used Flt3ligand 100ng/ml in RPMI1640 with 1*10 6 cell per ml. and then refresh half of the media on day 4 or 5 with fresh grow factor.

Hi Mohammad,

Our group seeds 1 - 1.5 x 106 cells/ml in 6-well plates, using 200ng/ml FLT3-L in RPMI supplemented with 10% FCS, 1% PenStrep, 0.1% 2-mercaptoethanol.

We culture for 7-9 days without exchanging the media. See K. Brasel, et al. Blood. 2000.
